DSS诱导果蝇溃疡性结肠炎模型的建立

葡聚糖硫酸钠(Dextran Sulfate Sodium,DSS)是葡聚糖的聚阴离子衍生物,喂食果蝇会破坏肠上皮中肠道表皮细胞之间的连接从而造成肠道损伤,引起多条通路应答调控干细胞的自我更新及分化,补充受损细胞,维持肠道稳态。

实验动物

5-10日龄果蝇,雌性 

 

实验材料

DSS(yeasen DSS 60316ES,MW36000-50000) 

 

实验步骤

1. 用5%的蔗糖溶液配制喂食培养基;
2. 对照组:5%蔗糖培养基(SC),实验组:3% DSS(培养基配制);
3. 预先用500 μL的喂食培养基将2.5cm×3.75cm的层析纸润湿,然后放入空的培养瓶中;
4. 果蝇放入含有层析纸的瓶中,29℃培养三天,期间每天将存活的果蝇移入含有新鲜培养基的空瓶中;
5. 培养三天后,切开中肠,用GFP(绿色)和PH3(红色)抗体和DRAQ5(蓝色)进行免疫染色。

 通过免疫染色中不同的标记方法,量化了肠道干细胞(ISC),ISC前体细胞发生增值,表明DSS诱导果蝇模型肠道发生损伤。
